Learning objectives: Lecture 13
Understand the different forms of chromatin (2nm, 11nm, 30nm fibres specifically)
Understand the structure of a nucleosome
List and understand the roles of the 5 histones and how they form to create a nucleosome
- And how H1 can create condensed DNA (and the effect of this on transcription)
Understand how and why the histone tails interact with each other, and also how they are modified
Understand the types and roles of:
- Histone modifying enzymes
- Chromodomain and bromodomain proteins, HATs, HDACs
- Chromatin modifying enzymes
- You should know the SWI/SNF enzymes but in a general sense (e.g. that they exist)
Understand how histone modification can be inherited during DNA replication
For proteins mentioned in the learning objectives, think about what would happen if you removed their functionality through mutagenesis.
Tools/assays to be familiar with and understand when and why they are used:
- DNAse hypersensitivity assay
- ChIP-seq (in a general sense - do understand what Co-IP is specifically, it is very similar to the ChIP in ChIP-seq)
