Learning objectives: Lecture 9
Understand the different outcome of a DNA nick or double stranded break (DSB).
Describe the four possible outcomes of a damaged template on the replication fork.
For the following mechanisms, understand/describe:
- The steps of the mechanism
- The proteins involved at each step
- The outcome(s) of the mechanism
Mechanisms to repair a DSB:
- Synthesis dependant strand annealing (SDSA)
- Homologous recombination
- Non homologous end joining (NHEJ)
Other mechanisms:
- Programmed generation of DSBs during meiosis
For mechanisms, think about what would happen if you removed the functionality of each of the proteins.
Assays to be familiar with:
- Shift/gel mobility assay
