Learning objectives: Lecture 9

Understand the different outcome of a DNA nick or double stranded break (DSB).

Describe the four possible outcomes of a damaged template on the replication fork.

For the following mechanisms, understand/describe:

  • The steps of the mechanism
  • The proteins involved at each step
  • The outcome(s) of the mechanism

Mechanisms to repair a DSB:

  • Synthesis dependant strand annealing (SDSA)
  • Homologous recombination
  • Non homologous end joining (NHEJ)

Other mechanisms:

  • Programmed generation of DSBs during meiosis

For mechanisms, think about what would happen if you removed the functionality of each of the proteins.

  • Shift/gel mobility assay
