Describe what a restriction endonuclease does and what they generate

Given a DNA sequence and a set of restriction endonucleases, be able to determine how the REs will digest the DNA

Define:

  • genomic library and how to create one
  • cDNA library and how to create one
  • probe

Describe the structure of a typical plasmid/vector

Describe the different ways we can transform bacteria

Describe the different types of vector (plasmid, BAC, YAC)
